Lethality and pathogenesis of airborne infection with filoviruses in A129 α/β -/- interferon receptor-deficient mice.
normal immunocompetent mice are not susceptible to the non-adjusted filoviruses. Therefore, there are two strategies available to build a model of murine infection Filovirus: adaptation of the virus to the host or the use of genetically modified mice that are susceptible to the virus. Some knockout (KO) strains of mice with defects either adaptive or innate immunity they are vulnerable to non-adjusted filoviruses.
In this study, A129 α / β – / – interferon receptor-deficient knockout mice, strain A129 IFN-α / β – / -, used to determine the lethal from various filoviruses, including Lake Victoria marburgvirus (Marv), Zaire ebolavirus (ZEBOV) , Sudan ebolavirus (SEBOV), Reston ebolavirus (REBOV) and Côte d’Ivoire ebolavirus (CIEBOV), administered using intraperitoneal (IP) or aerosol route of infection. One hundred percent mortality rate was observed in all groups given KO mice with different doses of Marv and ZEBOV challenge either by IP or aerosol route.
- While it was dead because it depends on both the dose and ranged from 5.4 to 7.4 days in the challenges of IP injection, and 10.2 to 13 days in aerosol challenge. Lethal dose (50% tissue culture infective dose, TCID (50)) of ZEBOV to KO mice was <1 TCID (50) ml (-1) when administered either by IP or aerosol route of infection; for Marv lethal dose was <1 TCID (50) ml (-1) with the IP route of infection and <10 TCID (50) ml (-1) via the aerosol route.
- In contrast, no mortality after infection with SEBOV or REBOV either by IP or aerosol route of infection; all rats lost weight (~ 15% loss of body weight means the group with SEBOV and ~ 7% with REBOV) but recovered to its original weight by day 14 post-challenge.
- There were no deaths in the mice given the CIEBOV via the IP route of infection and no clinical signs of infection were observed.
- More rapid disease progression after infection with ZEBOV than with Marv but eventually the two viruses cause widespread infection with high titer of infectious virus in some organs.
- Histopathological observations are consistent with other animal models and showed extensive organ damage. This study shows that Marv and ZEBOV more sinister when administered via the IP route instead by aerosol infection, although both highly virulent by one of the routes.
- KO mice could provide a useful model to test potential antiviral therapy against wild-type filoviruses.

Vesicular stomatitis virus-based Ebola vaccine by increasing the cross-protective efficacy.
For Ebola virus (EBOV), 4 different species are known: Zaire, Sudan, Ivory Coast, and Reston ebolavirus. The newly discovered Bundibugyo ebolavirus has been proposed as a species-5. So far, there is no cross-neutralization between EBOV species have been described, the progress of cross-species exacerbate protective vaccine.
By using the vesicular stomatitis virus (rVSV) based recombinant vaccines, guinea pigs can be protected against Zaire ebolavirus (ZEBOV) infection only when immunized with vectors expressing homologous, but not heterologous, EBOV glycoprotein (GP).
However, infection of guinea pigs with the wild type strain nonadapted of different species resulted in the full protection of all animals against subsequent challenge with guinea pig-adapted ZEBOV, indicating that cross-species protection is possible.
The resulting new vector containing EBOV virus protein 40 (VP40) or EBOV nucleoprotein (NP) as second antigen expressed by the same rVSV vectors encoding the heterologous GP. After applying the 2 dose immunization approach, we observed increased levels of cross-protection, with 5 of 6 guinea pigs live ZEBOV lethal challenge when vaccinated with rVSV-GP-expressing SEBOV and -VP40.
Our data indicate that cross-protection between species EBOV can be achieved, although EBOV-GP alone can not induce an immune response that is required.
